| In vitro study | G418 is an inhibitor of many prokaryotic and eukaryotic organisms at concentrations of 1-300 μg/ml. Resistance to G418 by the neo gene of Tn5 encoding aminoglycoside 3 '-phosphotransferase, APT 3'II is commonly used in laboratory studies to select genetically engineered cells [1]. Typically, for bacteria and algae a concentration of 5mg / L or less is used, for mammalian cells a concentration of about 400mg / L is used for selection and 200mg / L is used for maintenance. TRYPSIN INHIBITOR SOYBEAN REAGENT GRADE - Introduction
G418 is an aminoglycoside antibiotic. The structure of this aminoglycoside antibiotic is similar to neomycin, gentamicin, and kanamycin. In molecular genetic tests, it is the most commonly used resistance screening reagent for stable transfection. It blocks protein synthesis by inhibiting the genes of transposon Tn601,Tn5, interfering with ribosome function, and produces toxins to prokaryotic and eukaryotic cells, including bacteria, yeast, plant and mammalian cells, as well as protozoa and worms. When the neo gene is integrated into the eukaryotic cell DNA, it can initiate the transcription of the sequence encoded by the neo gene into mRNA, thereby obtaining the high-efficiency expression of the resistance product aminoglycoside phosphotransferase, so that the cell can obtain resistance and be able to contain G418 Growth in selective medium blocks polypeptide synthesis by interfering with ribosome function. Not a commonly used standard antibiotic, it is often used to select prokaryotic and eukaryotic cells transfected with neomycin resistance gene. | Animal experiment | In order to characterize the sensitivity of the trypanosome population to G418 in vivo, the blood flow forms of Brucella brucei GUTat 3.1 and Brucella brucei GUTat 3.1 / BBR3 were amplified separately in mice with sublethal radiation.
